Genetic Testing for Statin Induced Myopathy

چکیده
STATIN-INDUCED MYOPATHY Statins are associated with a known risk of muscle-related symptoms, which are the most common adverse effects of statin drugs. Myopathy is a general term for muscle toxicity. Three categories of statin-induced myopathy have been defined by a joint committee of the American College of Cardiology, American Heart Association, and National Heart, Lung and Blood Institute: Statin-induced myalgia, defined as any muscle symptoms that occur without an elevation of serum creatinine kinase (CK); Statin-induced myositis, defined as muscle symptoms with an elevation of serum CK; and Statin-induced rhabdomyolysis, defined as markedly severe muscle symptoms with an elevation of CK greater than 10 times normal with an elevation in serum creatinine.

Pharmacogenetics of Statin-Induced Myopathy: A Focused Review of the Clinical Translation of Pharmacokinetic Genetic Variants
Statins are the most commonly prescribed drugs in the United States and are extremely effective in reducing major cardiovascular events in the millions of Americans with hyperlipidemia. However, many patients (up to 25%) cannot tolerate or discontinue statin therapy due to statin-induced myopathy (SIM). Patients will continue to experience SIM at unacceptably high rates or experience unnecessar...
متن کاملCoenzyme Q(10) and statin myalgia: what is the evidence?
Statins lower cholesterol by inhibiting 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase, the rate-limiting enzyme in the biosynthesis of cholesterol. However, severe adverse events, including myalgias and rhabdomyolysis, have been reported with statin treatment. Different mechanisms have been proposed to explain statin-induced myopathy, including reduction of mevalonate pathway produc...
متن کاملElectrophysiologic and clinico-pathologic characteristics of statin-induced muscle injury
Objective(s):In this study, we aimed at evaluation of electrophysiological and histopathalogical characteristics of statin-induced muscle injury as well as clinical features of patients who develop this condition in terms of frequency and pattern of evolution. Materials and Methods: Forty patients (age 39-74 years) including 25 subjects with type 2 diabetes mellitus, 9 with cardiovascular disea...
متن کاملSLCO1B1 Genetic Variant Associated With Statin-Induced Myopathy: A Proof-of-Concept Study Using the Clinical Practice Research Datalink
This study aimed to determine whether patients with statin-induced myopathy could be identified using the United Kingdom Clinical Practice Research Datalink, whether DNA could be obtained, and whether previously reported associations of statin myopathy with the SLCO1B1 c.521T>C and COQ2 rs4693075 polymorphisms could be replicated. Seventy-seven statin-induced myopathy patients (serum creatine p...
متن کاملAutoimmune statin-induced myopathy: a case report
A 58-year-old woman with a history of statin use presented with a 4-month history of progressive weakness of both shoulders and thighs. Laboratory and electromyography testing confirmed the presence of generalized proximal myopathy and ruled out connective tissue disease, malignancy, or active viral infection. Muscle biopsy was consistent with necrotizing autoimmune myopathy.
